pout is not a standard MATLAB class, so pout.tif is not something we would expect to have defined.

Your two inputs appear to be the same (unless pout.tif is a method of a class that implements randomness.) The correlation coefficient will be 1.

Try this:
grayImage1 = double(imread('pout.tif'));
grayImage2 = grayImage1;
cc = corrcoef(grayImage1, grayImage2)
message = sprintf('The correlation coefficients are \n%f %f\n%f %f', cc)
helpdlg(message);
