Does MATLAB work on Windows 10 in "S mode"?

I hear the app store is very limited and no third party programs will be allowed.

It doesn't seem likely to me that Mathworks will develop a Windows 10 App, so your chances on getting Matlab to work on such a closed system probably aren't very good.
However, I might be wrong. The only people who can tell you for sure work for Mathworks. If you want an official answer, contact support directly (click on the phone icon at the top of the page).

The question is about Windows 10 S, a much more restricted version of Windows 10. Since S mode does not allow standalone applications, it's very unlikely that Matlab will ever work with it (see Rik's comment).

It is not garbage, it is just not for you. As far as I can tell it is for people who have some difficulty understanding how computers work. Windows 10S being so walled in makes it hard to mess up. If you can only install vetted apps from the store it will be impossible to install malware.
I can screen binaries I download myself. I can make sure I'm safe. I can re-install Windows if I royally mess up. My grandmother can't. 10S is terrible for me, it would probably be an improvement for her.
Windows 10 mode S is not aimed at individuals. It's mostly aimed at education, where schools supply computers to students with 10 S install, which gives the school much more control over which software is installed and stop the kids messing up the machine. Overall, a good idea for that market.
