intensity sum of smaller peak

hey all,

i have a series of images of reflected x-ray . these images contains one white with high intensity while another spot with less intensity exists at the same Y axis. I want to catch this small reflected spot having less intensity and get the sum of intensities in this smaller spot. can anybody help me out to write a code for this in MATLAB.
